Cartilage is a connective tissue found in many areas of the body including joints between bones (articular cartilage). Individuals whose cartilage is affected suffer from joint disease (arthritis) which is mainly degenerative and causes arthritis/osteoarthritis (OA) but also inflammatory arthritis including rheumatoid arthritis (RA) and ankylosing spondylitis (AS).

Bone turnover, also called bone remodelling, is a dynamic, lifelong process in which old bone is removed from the skeleton and new bone is added. The continual process of adult bone remodelling is essential for the maintenance of bone mass and skeletal micro-architecture. Osteoporosis is a disease characterised by low bone density and reduced structural strength of the bone leading to increased risk of bone fracture.

For the quantification of cartilage, degradation products of C-terminal telopeptides of type II collagen (CTX-II) assays are available for tissue culture supernatant, animal serum, urine and synovial fluid. Cartilage turnover can be investigated with cartilage oligomeric matrix protein (COMP) in serum and synovial fluid animal samples. Bone resorption C-terminal telopeptides (CTX-I) assays are available for cell culture, animal urine and serum samples.

Bone formation markers: Procollagen type I propeptides (PINP) is available for cell culture and rodent serum; Osteocalcin for serum samples (not in mice), and osteoclast marker (tartrate-resistant-acid phosphatase 5b (TRAcP5b)) are available as adequate assays for cell culture, and animal serum samples.
